Madekwe was born South London, England into a mixed family. Her mother is English and her father is Nigerian. Her family was raised on a South London community estate, and at the age of 14she enrolled in The Brit School. She studied acting in the Royal Academy of Dramatic Art, London. She appeared in many stages during her high time at school, including the King Henry V and Wuthering Heights.
Madekwe began her acting career with the movie Storm Damage. In the years since, she's made guest appearances in Teachers, Doctors, Casualty as well as Drop Dead Gorgeous. Madekwe made her debut motion picture in the 2007 Woody Allen film Cassandra's Dream, opposite Colin Farrell and Ewan McGregor. Following that success her role was Elisha in the single-episode BBC Three drama pilot West 10 LDN which aired in 2007 and she also appeared in six episodes of Trexx and Flipside in the role of Ollie. Madekwe starred as the lead character for How to Lose Friends And Alienate Others, a comedy made in 2008. Her credits in theatre include Little Sweet Thing by Roy Williams as well as Flight Path by David Watson.
